In this work, we characterize a complete, polymerization-competent building block of the C. elegans synaptonemal complex. Using mass photometry, crosslinking mass spectrometry, AlphaFold modeling, electron microscopy, and genetics, we identify the specific interfaces that mediate interactions within and between polymer units and validate them by mutagenesis both in vitro and in vivo.
